چکیده

ogists on the ABA-100, we found that about 13% of the samples (22 of 169) gave results which differed by more than 10 mg/liter. This prompted us to test the assumption concerning the effectiveness of the blanking wavelength. The test wavelength is 650 nm and the blank wavelength is 550 nm for the phosphorus procedure on the ABA-100. We Selected two patients’ samples that had given discrepant results and one that had not, and diluted them 26-fold with physiological saline. These solutions were scanned (vs. saline) from 700 to 400 nm with a ratio-recording spectrophotometer (DK-2A; Beckman Instruments, Fullerton, Calif. 92634). Table 1 shows the absorbance differences for these samples. The patients’ diagnoses were, respectively, cervical carcinoma, gunshot wound, and leukemia. All of these samples absorbed more at the blank wavelength than at the test wavelength and the absorbance difference varied with the patient. Results for phosphorus assays performed on these samples are also shown in the Table. These three examples serve to warn that extreme caution may have to be exercised whenever a bichnomatic analyzer is used for endpoint measurements. It may not be readily apparent how much comparison work is needed before a new procedure can be used with confidence. Rather, the amount of comparison work appears to be a function of the source(s) of samples that are being assayed by the particular laboratory; that is, of whether the patient population is relatively normal or is a combination of patients who are relatively normal and those who are receiving tertiary care. Another point also worth considening is that most of the method interferences documented to date are for procedures in which the blank and test mixtures are read at the same wavelength, information that may not be directly transferable to a bichromatic procedure. Fluorometric determination of "albumin-titratable bilirubin" in the jaundiced neonate.

We report a fluorometric technique for determination of albumin-titratable bilirubin in the jaundiced neonate. Although bilirubin alone has very little native fluorescence, considerable emission is observed in the presence of albumin under acid conditions. Analysis of the plasma sample alone and in the presence of excess human serum albumin solution appears to reflect the bilirubin tightly boun...

Reserve albumin binding capacity, salicylate saturation index, and red cell binding of bilirubin in neonatal jaundice.

Reserve albumin binding capacity, salicylate saturation index, and red cell binding of bili-rubin in neonatal jaundice. 105 blood samples from 72 infants, mostly with jaundice due to haemolytic disease, were analysed for reserve albumin binding capacity (HBABA method), salicylate saturation index (SI), and red cell binding of bilirubin. 2 infants with clinical symptoms of bilirubin encephalopat...

Glucocorticoids directly affect spectrophotometry of bilirubin in amniotic fluid.

Dexamethasone or prednisolone, added in vitro to bilirubin-containing amniotic fluid, produces a time-dependent decrease in the 450-nm absorbance of the pigment. Neither the chemical determination of bilirubin in amniotic fluid nor the lecithin/sphingomyelin ratio as determined by thin-layer chromatography is affected by these glucocorticoids. The effect probably is not a result of displacement...

Determination of unbound bilirubin in the serum of newborns.

An enzymatic assay is described for non-albuminbound bilirubin in the serum of newborn infants. Unbound bilirubin is oxidized to colorless compounds by ethyl hydroperoxide in the presence of horseradish peroxidase (EC 1.11.1.7), while albumin-bound bilirubin is protected from oxidation. Because the equilibrium between albumin and bilirubin occurs rapidly, the oxidation step is rate limiting, an...

Albumin-Bilirubin Binding Mechanism KINETIC AND SPECTROSCOPIC STUDIES OF BINDING OF BILIRUBIN AND XANTHOBILIRUBIC ACID TO HUMAN SERUM ALBUMIN*

After binding of bilirubin to human serum albumin (1:1), a train of relaxational changes of conformation takes place. The late part of these processes, occurring in the time interval 1-500 s , has been studied by recording the changes of light absorption. Similar processes have been demonstrated after binding of fatty acid anion to the bilirubin-albumin complex as well as after a pH-jump from 6...
